Declared Potency vs. Measured Enzyme Activity

Two numbers can appear on a supplement’s paperwork and look like the same fact. They are not. One is a commitment; the other is an observation. Confusing them is the most common way an enzyme label is misread — including by people acting in good faith.

The short answer

A declared specification is the activity level a product is formulated to meet — a label commitment, usually stated as a minimum. A measured result is what a laboratory assay actually found in a sample from one specific lot. A declared figure describes the product line. A measured figure describes a batch.

Declared: the specification

A declared specification is a formulation target and a label commitment. When a label states 250,000 SPU per capsule, that is the activity basis the product is built to, typically expressed as a minimum the lot must meet to be released.

A specification is meaningful. It defines what a lot must achieve, and it is the thing a release decision is made against. What it is not is a measurement of anything you own.

Measured: the assay result

A measured result comes from running the USP proteolytic assay on a sample from a specific lot: the enzyme is incubated with a protein substrate under controlled conditions, and the extent of protein breakdown is measured. The output is a number attached to that lot and no other.

Because enzyme activity varies with manufacturing, storage, and raw-material variability, that number is a fact about a moment and a batch. This is not a flaw in the testing — it is the reason per-lot testing exists at all.

Why the distinction is easy to lose

On a Certificate of Analysis, a specification column and a result column sit next to each other, and a document that restates the label figure in its results column has not reported a measurement. In marketing copy, the two collapse even faster: a specification can be described in language that sounds like a finding, without a single false word being written.

The test for the reader is simple. Ask: is this number what the product is supposed to contain, or what a laboratory found in a specific lot? The framework for answering that from the document itself is in How to Read a Serrapeptase Certificate of Analysis.

A labeled example: Lot 6798

Declared specification: at least 250,000 SPU per capsule. Measured result: 353,359 SPU.

That is Lot 6798’s own real, documented assay result. It is not a guarantee of what any other lot, past or future, will measure, and it should not be read as a typical figure, an average, or a product-wide claim. Every released lot is assayed and documented on its own terms, and a specific bottle’s result is retrievable by lot number through Veriphase™.

Why we label the example rather than feature the number

A single strong lot result is the easiest figure in this category to misuse. Presented without its lot, it becomes an implied promise about every bottle. Presented with its lot, it is what it actually is: one documented measurement, verifiable, and bounded.

What a measured result tells you

  • What one specific lot contained, by activity, at the point it was assayed.
  • Whether that lot met its declared specification.
  • That the specification is being verified rather than asserted — provided results are published per lot rather than once.

What it does not tell you

  • What any other lot measured. A lot result stays tied to its lot.
  • What the capsule does after you swallow it. Activity is measured in an assay, not in a person.
  • Whether the enzyme reaches the intestine intact. That is a delivery question, tested separately under USP <2040>.

Practical takeaway

When you encounter an activity figure, place it in one of two boxes before you use it: specification, or measurement. If it is a measurement, note which lot it belongs to. A number that cannot be placed in either box is not yet evidence.

A specification is a promise. A measured result is a receipt. Only one of them is about the bottle in your hand.
