What Does SPU Mean in Serrapeptase?

Serrapeptase labels almost always lead with a number followed by three letters: SPU. It is the single figure most often compared between products, and one of the least often explained. Understanding what the unit actually measures changes how you read every serrapeptase label you encounter.

The short answer

SPU stands for Serrapeptase Units. It is a measure of functional enzyme activity — how much enzymatic work a sample performs under standardized laboratory conditions — not a measure of how much material is in the capsule. Milligrams tell you weight. SPU tells you activity.

Activity is not the same as weight

Enzyme potency can be described two different ways: by mass, in milligrams of material, or by measured functional activity, meaning how much enzymatic work a given sample actually performs under standardized conditions. These are not interchangeable.

Two samples with identical milligram weights can differ substantially in how active they actually are, depending on manufacturing, storage, and raw-material variability. Enzymes are proteins, and a protein can be present by weight while having lost part of its functional capacity. This is why an activity unit exists at all: for this class of ingredient, weight alone does not describe what you are buying.

How the number is determined

Serrapeptase activity is determined via a USP proteolytic assay: a standardized laboratory test in which the enzyme is incubated with a protein substrate under controlled conditions of temperature, pH, and time, and the rate or extent of protein breakdown is measured, typically via a colorimetric readout. The result is expressed as units of activity per the tested quantity of material.

That design matters for a second reason. Because the assay measures a functional property specific to proteolytic activity of this type, a sample producing the expected activity signature under the assay also confirms that the material behaves as serrapeptase is expected to behave. The same test speaks to identity and to activity.

Per capsule or per daily dose?

This is the most common source of confusion when comparing labels, and it is not a technical subtlety — it changes the comparison entirely.

An SPU figure can be stated per capsule or per full daily serving. A product listing a larger number across two or three capsules may deliver less activity per capsule than one listing a smaller number for a single capsule. Before comparing two products, find the serving size on each label and normalize both figures to the same basis. Until you do, you are not comparing the same thing.

What the number tells you

  • The activity basis the product is formulated to, stated in a functional unit rather than a weight.
  • A figure you can normalize against other products, once you have confirmed the serving size each one refers to.
  • A property that can be independently measured, which means it can also be independently verified for a given lot.

What it does not tell you

  • What happens after you swallow the capsule. Activity measured in a laboratory assay describes the material tested, not absorption or any outcome in the body.
  • Whether the figure on the label is a declared specification or a measured result for the lot in your hand. Those are different statements — see Declared Potency vs. Measured Enzyme Activity.
  • Whether the enzyme is protected on the way through the stomach. That is a question about the delivery format, not the activity figure.

Practical takeaway

When you see an SPU figure, ask three questions in order: is it per capsule or per serving; is it a declared specification or a measured result; and can you see the document that measured it for the lot you actually own. A number without those three answers is a starting point, not a comparison.

SPU describes work performed under a defined test. Everything else you might want to know about a capsule is a separate question, answered by a separate test.
